One in six men will be diagnosed with PCa during their lifetime and about 217,000 new cases were diagnosed in 2010 in the United States.<a class="elsevierStyleCrossRef" href="#bib0005"><span class="elsevierStyleSup">1</span></a> Several epidemiologic and molecular pieces of evidence suggest that inflammation is an important component in the etiology of PCa. However, the role of inflammation in the development of PCa is still unclear.</p><p id="par0010" class="elsevierStylePara elsevierViewall">Myeloperoxidase (MPO) is recruited and released as part of the inflammation response, which is an endogenous oxidant enzyme that generates reactive oxygen species (ROS) and can be released outside the cell, raising the potential for damage to an extracellular target.<a class="elsevierStyleCrossRef" href="#bib0010"><span class="elsevierStyleSup">2</span></a> MPO has been indirectly implicated in playing a role in carcinogenesis through both activation of procarcinogens to genotoxic intermediates and the potentiation of xenobiotic carcinogenicity.<a class="elsevierStyleCrossRefs" href="#bib0015"><span class="elsevierStyleSup">3,4</span></a></p><p id="par0015" class="elsevierStylePara elsevierViewall">The human MPO gene is located on chromosome 17q23.1 and consists of 12 exons and 11 introns. Two recently described single-nucleotide polymorphisms (SNPs) in the promoter region of the MPO gene can potentially affect MPO gene transcription and protein levels, but only limited data are available to support this contention.<a class="elsevierStyleCrossRefs" href="#bib0025"><span class="elsevierStyleSup">5,6</span></a> Besides, William et al. reported that the substitution of thymidine for cytosine in exon 10 at codon 569 caused arginine (CGG) changes to tryptophan (TGG), which might cause genetic defects of MPO.<a class="elsevierStyleCrossRef" href="#bib0035"><span class="elsevierStyleSup">7</span></a></p><p id="par0020" class="elsevierStylePara elsevierViewall">In our study, we genotyped all the known common SNPs within the genomic region of MPO (including 13 SNPs of the gene) from HapMap and evaluated their associations between the polymorphisms and PCa risk in a hospital-based case–control study in a population of Han nationality.</p></span><span id="sec0010" class="elsevierStyleSection elsevierViewall"><span class="elsevierStyleSectionTitle">Material and methods</span><span id="sec0015" class="elsevierStyleSection elsevierViewall"><span class="elsevierStyleSectionTitle">Study subjects</span><p id="par0025" class="elsevierStylePara elsevierViewall">The study subjects consisted of 1108 cases with newly diagnosed PCa and 1525 cancer-free male controls, which has been described previously.<a class="elsevierStyleCrossRefs" href="#bib0040"><span class="elsevierStyleSup">8,9</span></a> All cases were diagnosed with PCa through biopsy of puncture (ultrasound-guided transrectal needle biopsy of prostate, >10-fold biopsy) or operation (radical prostatectomy and transurethral resection of the prostate). Pathological, serological (prostate specific antigen, prostatic acid phosphatase), physical and other auxiliary examinations were performed on all controls in order to exclude the possibility of PCa. All subjects included in this study were ethnic Han Chinese, permanently residing in China, which were previously described in detail. Each subject was informed about the aims and requirements of this study, and informed consent for participation was obtained in accordance with institutional guidance at Fudan University, the Shanghai Cancer Institute, and each participating hospital.</p></span><span id="sec0020" class="elsevierStyleSection elsevierViewall"><span class="elsevierStyleSectionTitle">Genotyping</span><p id="par0030" class="elsevierStylePara elsevierViewall">A total of 13 SNPs of MPO gene were selected from HapMap for genotyping. Samples from all 2633 subjects included in this study were genotyped on the MassARRAY iPLEX (Sequenom, Inc., San Diego, CA) platform. Technicians that performed the genotyping were blinded to the case–control status of all subjects.</p></span><span id="sec0025" class="elsevierStyleSection elsevierViewall"><span class="elsevierStyleSectionTitle">Statistical analysis</span><p id="par0035" class="elsevierStylePara elsevierViewall">All differences in selected SNPs between the case and control groups were evaluated by using Pearson's Chi-squared tests for Hardy–Weinberg equilibrium. Unconditional univariate and multivariate logistic regression analyses were performed to obtain the crude and adjusted odds ratios (ORs) for the risk of PCa and their 95% confidence intervals (CIs). All analyses were performed using Statistical Analysis System (SAS) software (version 9.2; SAS Institute, Cary, NC, USA).</p></span></span><span id="sec0030" class="elsevierStyleSection elsevierViewall"><span class="elsevierStyleSectionTitle">Results</span><p id="par0040" class="elsevierStylePara elsevierViewall">The characteristics of the 1108 cases and 1525 controls were previously shown in detail.<a class="elsevierStyleCrossRef" href="#bib0040"><span class="elsevierStyleSup">8</span></a> We evaluated 13 SNPs in MPO gene and their association with prostate cancer risk (<a class="elsevierStyleCrossRef" href="#tbl0005">Table 1</a>). We found that only one SNP (rs8082134, <span class="elsevierStyleItalic">P</span><span class="elsevierStyleHsp" style=""></span>=<span class="elsevierStyleHsp" style=""></span>0.02286) did differ significantly between two groups but the OR of this SNP was 0.221. Another SNP showed a limited association with PCa risk (rs7208693, <span class="elsevierStyleItalic">P</span><span class="elsevierStyleHsp" style=""></span>=<span class="elsevierStyleHsp" style=""></span>0.0877, OR<span class="elsevierStyleHsp" style=""></span>=<span class="elsevierStyleHsp" style=""></span>0.8745). But other SNPs of MPO had no significant difference between genotype distributions in the PCa and control groups.</p><elsevierMultimedia ident="tbl0005"></elsevierMultimedia></span><span id="sec0035" class="elsevierStyleSection elsevierViewall"><span class="elsevierStyleSectionTitle">Discussion</span><p id="par0045" class="elsevierStylePara elsevierViewall">MPO is an enzyme that mediated generation of hydroxy radicals in the presence of superoxide.<a class="elsevierStyleCrossRef" href="#bib0050"><span class="elsevierStyleSup">10</span></a> Its reactive by-products have been associated with DNA strand breakage, DNA base modification, inhibition of DNA repair, and generation of carcinogens.<a class="elsevierStyleCrossRefs" href="#bib0055"><span class="elsevierStyleSup">11–14</span></a></p><p id="par0050" class="elsevierStylePara elsevierViewall">However, there were few researches on the association between the SNPs of MPO and PCa risk in the Chinese Han population. Many SNPs, which were in linkage disequilibrium (LD) with other nearby SNPs in the genome, make it so feasible for us to select a small number of genetic markers, also called tagging single-nucleotide polymorphisms (tSNPs), to find the common variations of a gene. Because tSNPs provide information about nearby SNPs that were not genotyped, the use of tSNPs could improve the effectiveness of association research.<a class="elsevierStyleCrossRef" href="#bib0075"><span class="elsevierStyleSup">15</span></a> The International HapMap Project was a great resource that provided genome-wide data to support such tSNP analyses.<a class="elsevierStyleCrossRef" href="#bib0080"><span class="elsevierStyleSup">16</span></a> In our study, we selected 13 SNPs from the data for the Chinese in the HapMap (<a href="http://www.hapmap.%20org/">http://www.hapmap. org/</a>) to assess the association between common variations of genes in MPO and risk of prostate cancer in our ongoing hospital-based case–control study in a Chinese population.</p><p id="par0055" class="elsevierStylePara elsevierViewall">In our research, we found that the SNP rs8082134 was significant. There was no report about this SNP. It was an intronic polymorphism which had been reported to be associated with a variety of chronic diseases including breast cancer,<a class="elsevierStyleCrossRef" href="#bib0085"><span class="elsevierStyleSup">17</span></a> essential hypertension,<a class="elsevierStyleCrossRef" href="#bib0090"><span class="elsevierStyleSup">18</span></a> and type II diabetes.<a class="elsevierStyleCrossRef" href="#bib0095"><span class="elsevierStyleSup">19</span></a> However, its functional consequences were less known. Recently, data from immunoprecipitation and gene expression experiments showed that up to 40% of transcription factor binding sites were located within introns.<a class="elsevierStyleCrossRef" href="#bib0100"><span class="elsevierStyleSup">20</span></a> But the OR of this SNP was 0.2212, so we did not think it had an association with the susceptibility to PCa in the Chinese population.</p><p id="par0060" class="elsevierStylePara elsevierViewall">Besides, another SNP that also caused our interest was rs7208693 (<span class="elsevierStyleItalic">P</span><span class="elsevierStyleHsp" style=""></span>=<span class="elsevierStyleHsp" style=""></span>0.0877, OR<span class="elsevierStyleHsp" style=""></span>=<span class="elsevierStyleHsp" style=""></span>0.8745), though it did not show a significant difference between the PCa and control groups. This was similar to those found by Sarah et al.<a class="elsevierStyleCrossRef" href="#bib0105"><span class="elsevierStyleSup">21</span></a> and Sun et al.<a class="elsevierStyleCrossRef" href="#bib0110"><span class="elsevierStyleSup">22</span></a> The C>A polymorphisms of rs7208693 can also lead to the changing of phenylalanine (Phe) on the No. 53 of MPO enzyme into valine (Val).</p></span><span id="sec0040" class="elsevierStyleSection elsevierViewall"><span class="elsevierStyleSectionTitle">Conclusion</span><p id="par0065" class="elsevierStylePara elsevierViewall">In summary, one SNP of MPO was significant, but our study revealed that other genotype frequencies of MPO gene polymorphisms do not vary between PCa patients and control subjects in the Chinese population. This result shows that these polymorphisms had limited effect in PCa.
