still ranks high as the leading cause of death among urological malignancies&#46; One in six men will be diagnosed with PCa during their lifetime and about 217&#44;000 new cases were diagnosed in 2010 in the United States&#46;<a class="elsevierStyleCrossRef" href="#bib0005"><span class="elsevierStyleSup">1</span></a> Several epidemiologic and molecular pieces of evidence suggest that inflammation is an important component in the etiology of PCa&#46; However&#44; the role of inflammation in the development of PCa is still unclear&#46;</p><p id="par0010" class="elsevierStylePara elsevierViewall">Myeloperoxidase &#40;MPO&#41; is recruited and released as part of the inflammation response&#44; which is an endogenous oxidant enzyme that generates reactive oxygen species &#40;ROS&#41; and can be released outside the cell&#44; raising the potential for damage to an extracellular target&#46;<a class="elsevierStyleCrossRef" href="#bib0010"><span class="elsevierStyleSup">2</span></a> MPO has been indirectly implicated in playing a role in carcinogenesis through both activation of procarcinogens to genotoxic intermediates and the potentiation of xenobiotic carcinogenicity&#46;<a class="elsevierStyleCrossRefs" href="#bib0015"><span class="elsevierStyleSup">3&#44;4</span></a></p><p id="par0015" class="elsevierStylePara elsevierViewall">The human MPO gene is located on chromosome 17q23&#46;1 and consists of 12 exons and 11 introns&#46; Two recently described single-nucleotide polymorphisms &#40;SNPs&#41; in the promoter region of the MPO gene can potentially affect MPO gene transcription and protein levels&#44; but only limited data are available to support this contention&#46;<a class="elsevierStyleCrossRefs" href="#bib0025"><span class="elsevierStyleSup">5&#44;6</span></a> Besides&#44; William et al&#46; reported that the substitution of thymidine for cytosine in exon 10 at codon 569 caused arginine &#40;CGG&#41; changes to tryptophan &#40;TGG&#41;&#44; which might cause genetic defects of MPO&#46;<a class="elsevierStyleCrossRef" href="#bib0035"><span class="elsevierStyleSup">7</span></a></p><p id="par0020" class="elsevierStylePara elsevierViewall">In our study&#44; we genotyped all the known common SNPs within the genomic region of MPO &#40;including 13 SNPs of the gene&#41; from HapMap and evaluated their associations between the polymorphisms and PCa risk in a hospital-based case&#8211;control study in a population of Han nationality&#46;</p></span><span id="sec0010" class="elsevierStyleSection elsevierViewall"><span class="elsevierStyleSectionTitle">Material and methods</span><span id="sec0015" class="elsevierStyleSection elsevierViewall"><span class="elsevierStyleSectionTitle">Study subjects</span><p id="par0025" class="elsevierStylePara elsevierViewall">The study subjects consisted of 1108 cases with newly diagnosed PCa and 1525 cancer-free male controls&#44; which has been described previously&#46;<a class="elsevierStyleCrossRefs" href="#bib0040"><span class="elsevierStyleSup">8&#44;9</span></a> All cases were diagnosed with PCa through biopsy of puncture &#40;ultrasound-guided transrectal needle biopsy of prostate&#44; &#62;10-fold biopsy&#41; or operation &#40;radical prostatectomy and transurethral resection of the prostate&#41;&#46; Pathological&#44; serological &#40;prostate specific antigen&#44; prostatic acid phosphatase&#41;&#44; physical and other auxiliary examinations were performed on all controls in order to exclude the possibility of PCa&#46; All subjects included in this study were ethnic Han Chinese&#44; permanently residing in China&#44; which were previously described in detail&#46; Each subject was informed about the aims and requirements of this study&#44; and informed consent for participation was obtained in accordance with institutional guidance at Fudan University&#44; the Shanghai Cancer Institute&#44; and each participating hospital&#46;</p></span><span id="sec0020" class="elsevierStyleSection elsevierViewall"><span class="elsevierStyleSectionTitle">Genotyping</span><p id="par0030" class="elsevierStylePara elsevierViewall">A total of 13 SNPs of MPO gene were selected from HapMap for genotyping&#46; Samples from all 2633 subjects included in this study were genotyped on the MassARRAY iPLEX &#40;Sequenom&#44; Inc&#46;&#44; San Diego&#44; CA&#41; platform&#46; Technicians that performed the genotyping were blinded to the case&#8211;control status of all subjects&#46;</p></span><span id="sec0025" class="elsevierStyleSection elsevierViewall"><span class="elsevierStyleSectionTitle">Statistical analysis</span><p id="par0035" class="elsevierStylePara elsevierViewall">All differences in selected SNPs between the case and control groups were evaluated by using Pearson&#39;s Chi-squared tests for Hardy&#8211;Weinberg equilibrium&#46; Unconditional univariate and multivariate logistic regression analyses were performed to obtain the crude and adjusted odds ratios &#40;ORs&#41; for the risk of PCa and their 95&#37; confidence intervals &#40;CIs&#41;&#46; All analyses were performed using Statistical Analysis System &#40;SAS&#41; software &#40;version 9&#46;2&#59; SAS Institute&#44; Cary&#44; NC&#44; USA&#41;&#46;</p></span></span><span id="sec0030" class="elsevierStyleSection elsevierViewall"><span class="elsevierStyleSectionTitle">Results</span><p id="par0040" class="elsevierStylePara elsevierViewall">The characteristics of the 1108 cases and 1525 controls were previously shown in detail&#46;<a class="elsevierStyleCrossRef" href="#bib0040"><span class="elsevierStyleSup">8</span></a> We evaluated 13 SNPs in MPO gene and their association with prostate cancer risk &#40;<a class="elsevierStyleCrossRef" href="#tbl0005">Table 1</a>&#41;&#46; We found that only one SNP &#40;rs8082134&#44; <span class="elsevierStyleItalic">P</span><span class="elsevierStyleHsp" style=""></span>&#61;<span class="elsevierStyleHsp" style=""></span>0&#46;02286&#41; did differ significantly between two groups but the OR of this SNP was 0&#46;221&#46; Another SNP showed a limited association with PCa risk &#40;rs7208693&#44; <span class="elsevierStyleItalic">P</span><span class="elsevierStyleHsp" style=""></span>&#61;<span class="elsevierStyleHsp" style=""></span>0&#46;0877&#44; OR<span class="elsevierStyleHsp" style=""></span>&#61;<span class="elsevierStyleHsp" style=""></span>0&#46;8745&#41;&#46; But other SNPs of MPO had no significant difference between genotype distributions in the PCa and control groups&#46;</p><elsevierMultimedia ident="tbl0005"></elsevierMultimedia></span><span id="sec0035" class="elsevierStyleSection elsevierViewall"><span class="elsevierStyleSectionTitle">Discussion</span><p id="par0045" class="elsevierStylePara elsevierViewall">MPO is an enzyme that mediated generation of hydroxy radicals in the presence of superoxide&#46;<a class="elsevierStyleCrossRef" href="#bib0050"><span class="elsevierStyleSup">10</span></a> Its reactive by-products have been associated with DNA strand breakage&#44; DNA base modification&#44; inhibition of DNA repair&#44; and generation of carcinogens&#46;<a class="elsevierStyleCrossRefs" href="#bib0055"><span class="elsevierStyleSup">11&#8211;14</span></a></p><p id="par0050" class="elsevierStylePara elsevierViewall">However&#44; there were few researches on the association between the SNPs of MPO and PCa risk in the Chinese Han population&#46; Many SNPs&#44; which were in linkage disequilibrium &#40;LD&#41; with other nearby SNPs in the genome&#44; make it so feasible for us to select a small number of genetic markers&#44; also called tagging single-nucleotide polymorphisms &#40;tSNPs&#41;&#44; to find the common variations of a gene&#46; Because tSNPs provide information about nearby SNPs that were not genotyped&#44; the use of tSNPs could improve the effectiveness of association research&#46;<a class="elsevierStyleCrossRef" href="#bib0075"><span class="elsevierStyleSup">15</span></a> The International HapMap Project was a great resource that provided genome-wide data to support such tSNP analyses&#46;<a class="elsevierStyleCrossRef" href="#bib0080"><span class="elsevierStyleSup">16</span></a> In our study&#44; we selected 13 SNPs from the data for the Chinese in the HapMap &#40;<a href="http://www.hapmap.%20org/">http&#58;&#47;&#47;www&#46;hapmap&#46; org&#47;</a>&#41; to assess the association between common variations of genes in MPO and risk of prostate cancer in our ongoing hospital-based case&#8211;control study in a Chinese population&#46;</p><p id="par0055" class="elsevierStylePara elsevierViewall">In our research&#44; we found that the SNP rs8082134 was significant&#46; There was no report about this SNP&#46; It was an intronic polymorphism which had been reported to be associated with a variety of chronic diseases including breast cancer&#44;<a class="elsevierStyleCrossRef" href="#bib0085"><span class="elsevierStyleSup">17</span></a> essential hypertension&#44;<a class="elsevierStyleCrossRef" href="#bib0090"><span class="elsevierStyleSup">18</span></a> and type II diabetes&#46;<a class="elsevierStyleCrossRef" href="#bib0095"><span class="elsevierStyleSup">19</span></a> However&#44; its functional consequences were less known&#46; Recently&#44; data from immunoprecipitation and gene expression experiments showed that up to 40&#37; of transcription factor binding sites were located within introns&#46;<a class="elsevierStyleCrossRef" href="#bib0100"><span class="elsevierStyleSup">20</span></a> But the OR of this SNP was 0&#46;2212&#44; so we did not think it had an association with the susceptibility to PCa in the Chinese population&#46;</p><p id="par0060" class="elsevierStylePara elsevierViewall">Besides&#44; another SNP that also caused our interest was rs7208693 &#40;<span class="elsevierStyleItalic">P</span><span class="elsevierStyleHsp" style=""></span>&#61;<span class="elsevierStyleHsp" style=""></span>0&#46;0877&#44; OR<span class="elsevierStyleHsp" style=""></span>&#61;<span class="elsevierStyleHsp" style=""></span>0&#46;8745&#41;&#44; though it did not show a significant difference between the PCa and control groups&#46; This was similar to those found by Sarah et al&#46;<a class="elsevierStyleCrossRef" href="#bib0105"><span class="elsevierStyleSup">21</span></a> and Sun et al&#46;<a class="elsevierStyleCrossRef" href="#bib0110"><span class="elsevierStyleSup">22</span></a> The C&#62;A polymorphisms of rs7208693 can also lead to the changing of phenylalanine &#40;Phe&#41; on the No&#46; 53 of MPO enzyme into valine &#40;Val&#41;&#46;</p></span><span id="sec0040" class="elsevierStyleSection elsevierViewall"><span class="elsevierStyleSectionTitle">Conclusion</span><p id="par0065" class="elsevierStylePara elsevierViewall">In summary&#44; one SNP of MPO was significant&#44; but our study revealed that other genotype frequencies of MPO gene polymorphisms do not vary between PCa patients and control subjects in the Chinese population&#46;
