Introducción

El CD44 es una molécula de adhesión celular de la que se conocen una forma estándar (CD44s) y varias isoformas variantes (CD44v) y cuyo principal ligando es el ácido hialurónico (AH), proteglicano de la matriz extracelular implicado en numerosas acciones biológicas. En este estudio hemos querido analizar la posible influencia de la concentración de AH sobre ciertas propiedades clinicobiológicas de los carcinomas ductales infiltrantes de mama CD44v5 positivos.

Pacientes y métodos

Se han determinado en 124 carcinomas ductales infiltrantes de mama CD44v5 positivos (56 AH positivos [> 1.500 ng/mg de proteína] y 68 AH negativos) las concentraciones citosólicas de receptores de estrógenos (RE), de progesterona (RP), ps2, catepsina D y activador del plasminógeno tipo tisular (t-AP). Se ha considerado también el tamaño tumoral, afección ganglionar axilar (N), el grado histológico (GH), ploidía y la fase de síntesis celular (FS).

Resultados

Los tumores AH positivos presentaron menores concentraciones de catepsina D (p = 0,013) que los AH negativos. Asimismo, fueron menos frecuentemente aneuploides (p = 0,026), catepsina D positivos (p = 0,015), GH 3 (p = 0,043) y proliferativos (FS > 7%; p = 0,049).

Conclusiones

Los resultados anteriores sugieren que la concentración de ácido hialurónico en la membrana celular podría asociarse con ciertas propiedades clinicobiológicas de los carcinomas ductales infiltrantes de mama CD44v5 positivos.

Introduction

CD44 is a cell adhesion molecule. There is one standard form, CD44s, and many variant isoforms, CD44v; their main ligand is hyaluronic acid, which is a proteoglycan found in the extracellular matrix and is involved in various biological functions. In this study we analyzed whether hyaluronic acid levels in the cell membrane modulated certain clinical and biological features of CD44v5-positive infiltrating ductal carcinomas of the breast.

Patients and methods

We determined 124 patients with CD44v5-positive infiltrating ductal carcinoma of the breast. Of these, 56 were hyaluronic positive (> 1,500 ng/mg prot.) and 68 were negative. We measured the cytoplasmic levels of oestrogen receptors, progesterone receptors, ps2, cathepsin D, and tissue-type plasminogen activator. Tumour size, axillary lymph node involvement, histological grade, ploidy and Sphase of cell cycle were also taken into account.

Results

Hyaluronic positive tumors had lower concentrations of cathepsin D (p = 0.013) than hyaluronic negative tumors. They were also less frequently aneuploid (p = 0.026) and cathepsin D positive (p = 0.015), histological grade 3 (p = 0.043) and S-phase > 7% (p = 0.049).

Conclusions

Our results suggest that hyaluronic acid concentrations in the cell membrane may be associated with some clinical and biological differences between CD44v5-positive infiltrating ductal carcinomas of the breast.
