Objetivo

Estudiar el efecto del ácido araquidónico (AA) y la implicación de la fosfolipasa-A2 (PLA2) sobre la función de barrera y la secreción electrogénica de cloro en el epitelio intestinal.

Material y métodos

Se llevaron a cabo estudios electrofisiológicos en la línea T84 y en el colon distal de rata. Se determinaron la secreción iónica (Isc) y la resistencia transepitelial (RT).

Resultados

El AA exógeno potenció la secreción estimulada por calcio en la línea T84. La incubación del colon distal de rata en presencia de ácido aristolóquico, un inhibidor de la PLA2, disminuyó la Isc basal sin alterar la RT e inhibió la Isc estimulada por un agonista del AMPc, la forscolina. En células T84, el ácido aristolóquico inhibió la secreción estimulada tanto por carbacol como por forscolina.

Conclusión

La PLA2 modula la secreción electrogénica de cloro, sin ejercer ningún efecto sobre la función de barrera en células de la línea T84 y en colon distal de rata.

Aim

To study the effect of aristolochic acid (AA) and the effect of phospholipase A2 (PLA2) on barrier function and electrogenic chloride secretion in intestinal epithelium.

Material and Methods

Electrophysiological studies were performed in the T84 cell line and rat distal colon. Ionic secretion and transepithelial resistance were determined.

Results

Exogenous AA increased calcium-stimulated secretion in the T84 cell line. Incubation of rat distal colon in the presence of AA, a PLA2 inhibitor, reduced basal ionic secretion without affecting transepithelial resistance and inhibited ionic secretion stimulated by a cyclic adenosine monophosphate (AMPc) agonist, forskolin. In T84 cells, AA inhibited both carbachol-and forskolin-stimulated secretion.

Conclusion

PLA2 modulates electrogenic chloride secretion but has no effect on barrier function in the T84 cell line or in rat distal colon.
